Grindelwald


今日はジャーナルクラブで発表の番。ということで、昨晩ちょっと遅くまで準備して紹介してきたのは以下の論文。

The Drosophila TNF receptor Grindelwald couples loss of cell polarity and neoplastic growth.  Nature, doi:10.1038/nature14298.
Disruption of epithelial polarity is a key event in the acquisition of neoplastic growth.  JNK signalling is known to play an important part in driving the malignant progression of many epithelial tumours, although the link between loss of polarity and JNK signalling remains elusive.  In a Drosophila genome-wide genetic screen designed to identify molecules implicated in neoplastic growth, we identified grindelwald (grnd), a gene encoding a transmembrane protein with homology to members of the tumour necrosis factor receptor (TNFR) superfamily.  Here we show that Grnd mediates the pro-apoptotic functions of Eiger (Egr), the unique Drosophila TNF, and that overexpression of an active form of Grnd lacking the extracellular domain is sufficient to activate JNK signalling in vivo.  Grnd also promotes the invasiveness of RasV12/scrib-/- tumours through Egr-dependent Matrix metalloprotease-1 (Mmp1) expression.  Grnd localizes to the subapical membrane domain with the cell polarity determinant Crumbs (Crb) and couples Crb-induced loss of polarity with JNK activation and neoplastic growth through physical interaction with Veli (also known as Lin-7).  Therefore, Grnd represents the first example of a TNFR that integrates signals from both Egr and apical polarity determinants to induce JNK-dependent cell death or tumour growth.

3年ほど前に、developmental timingを制御する「Dilp8」を報告したニースのDr. PL labからの論文。その3年前の論文で行われたRNAi screeningでは、imaginal discにneoplastic tumorが誘導される rn>avl-RNAi をtester lineとして、10100 RNAi lines中の121 linesでpupariation delayがレスキューされたとしている。このとき、slower disc growthが誘導される rn>rpl7-RNAi をtester lineとして行われたsecond screeningでかかってきたのが Dilp8。で、今回の論文ではそのfirst screeningでかかってきた121 linesの中で、avl knockdownによるneoplastic overgrowthがレスキューされていた8 linesに注目。そのうちの5つはJNK pathwayのcore components。で、残りのうち一つが今回Drosophila TNF receptorとして報告された「Grindelwald (Grnd)」である。あと二つがなんなのか気になるね。
Grindelwaldとは、スイスのベルナーオーバーラントにある村の名前であり、オーバーラント三山の一つ Eiger の麓にある。もちろん、Drosophila TNFである Eiger (Egr) のレセプターとして発見されたからGrindelwaldと名付けられたわけだが、同様の理由で以前に「Wengen (Wgn)」(こちらもEigerの麓にある村の名前)と名付けられたTNFRが存在する。ところがこの論文では、Egr-induced apoptosisにおいてTNFRとして機能しているのはWgnではなくGrndであるとして、以前に報告されているデータとの矛盾を指摘している。そういう意味でちょっと話題になっていた印象もあるけど、この論文で一番大事なのは、このGrndがapical polarity determinantであるCrumbs complexとJNK pathwayをmediateしているということやね。今まで、nTSG mutantのようなapicobasal polarity defectを持つ細胞が、JNK-dependent cell death or tumor growthを示すことは何度も確かめられてきたけども、なぜそのような細胞でJNK activationが起こるのかは明確にされてこなかった。Egr-JNK signaling by endocytic activationとか、Egr expression by tumor-associated hemocytesとかいう話はあるんやけどね。今これを書いていて思い出したけど、6年前にバルセロナのSpeakers Dinnerの席でまさにこの「How is JNK activated in nTSG mutant cells?」というquestionについて、Dr. EMとDr. GHとDr. MVの四人でワインを飲みながら真剣に話したんだった。
それにしてもやっぱり未だにスッキリせん部分があるよね。この論文でも結局は"context dependent”というように、cell competitionのようなときにはJNK activationがapoptosisを誘導するけど、homogeneous environmentではJNK activationがtumor growthを促すのはなんでなんやろうか。まぁこれはJNKのdownstreamで何が選択されるのかという部分がcontext dependentになるんかな。
avl-knockdowned cellsではapical hypertrophyが起こっていて、CrbとGrndのapical localizationがexpandしている。これは以下、Hippo signalingをdisturbしてYki activation、cell proliferationと繋がるんよね。でもどうやらこちらのcascadeにGrndは関わっていないようだと。それはエエんやけど、GrndがPDZ domainを介してVeliとinteractしているとしても、なんでGrnd knockdownでtissue disorganizationのほうがレスキューされるんかな。まぁ、JNKをブロックしても同様なわけだから、これもJNK downstreamにおける何が重要なのかという話になるのか。。。

とかそんなことを考えているうちにバルセロナへの出発が一週間後に迫ってきた。あんまり考えないでこの論文を選んだけど、なんだか "Cancer Modelling in Drosophila” なモードになってきたので良かったかな。
